Necessity of Liquid Nitrogen Refrigeration and Grinding
In molecular biology experiments, animal muscle tissue, connective tissue, bone tissue, hair tissue, plant roots, stems, leaves, seeds and so on need to be ground, and then its composition, gene and protein, structure and function are studied.
In order to make all kinds of tissue components not easy to be destroyed or degraded in the grinding process, but also to make the tissue hard and brittle, so that it is easy to grind, especially for the grinding separation and extraction of RNA and DNA. It is necessary to put biomaterials in liquid nitrogen for quick freezing to terminate all biological reactions inside and outside the cell. At the same time, the biomaterial cells are completely frozen and brittle, so that grinding can achieve good Cell-breaking effect, grinding the cells into powder and releasing the substances inside.

Liquid Nitrogen Refrigeration Grinding Procedure
The tissue samples were subdivided into suitable centrifugal tubes.
Adding appropriate amount of pyrolysis medium in centrifugal tube
Tighten the cap of the centrifuge and insert it into the freezing adapter.
Using special adapter fixture, the refrigeration adapter is placed in liquid nitrogen for 3-15 minutes.
Then use the special adapter fixture, put the freezing adapter out, put it into Gd16 plus grinder, and tighten the fixed handle.
Set up grinding speed and time, start Gd16 plus grinding instrument
After the grinder stops, the refrigeration adapter is removed and the centrifugal tube is removed.
Matters needing attention
The temperature of liquid nitrogen at atmospheric pressure is quite low, so extra attention should be paid to the use of liquid nitrogen to avoid direct contact with skin.
2. To place the freezing adapter in liquid nitrogen and take it out of liquid nitrogen, special adapter fixture should be used and anti-freezing gloves should be worn.
